Description The BRCA-1 gene codes for a nuclear phosphoprotein that plays a role in maintaining genomic stability and acts as a tumor suppressor. The normal gene plays a role in repairing breaks in DNA. If a mutation occurs in this gene the repair function may become disabled thus leading to more DNA replication errors and neoplastic growth. Current findings suggest that BRCA-1 may play an as yet undefined protective role in cells, as it is strongly expressed in epithelial cells undergoing high levels of proliferation in association with differentiation. Description C1q, a subcomponent of the classical complement pathway, is composed of nine subunits that mediate classical complement activation and thereby play an important role in the immune response. C1q associates with the proenzymes C1r and C1s to yield C1, the first component of the serum complement system. The collagen-like regions of C1q interact with the Ca(2+)-dependent C1r(2)C1s(2) proenzyme complex, and efficient activation of C1 takes place on interaction of the globular heads of C1q with the Fc regions of IgG or IgM antibody present in immune complexes. Description Complement component C3 plays a central role in the activation of complement system. Its activation is required for both classical and alternative complement activation pathways. C3d deposition in the renal transplant PTCs (peritubular capillaries) is indicative of AR (acute rejection) with subsequent high probability of graft loss. Anti-C3d, combined with anti-C4d, can be utilized as a tool for diagnosis of AR and warrant prompt and aggressive anti-rejection treatment. It was shown that anti-C3d labeled the epidermal basement membrane in 97% (31/32) cases of bullous pemphigoid (BP), with none of the normal controls demonstrating such findings. In the same study 27% (3/11) cases of pemphigus vulgaris (PV) demonstrated intercellular C3d deposition. Therefore, C3d immunohistochemistry is a helpful adjunct in the diagnosis of BP (and perhaps PV), especially in the cases in which only formalin-fixed, paraffin embedded tissue is available for analysis.
