PDLIM1 (PDZ and LIM domain 1), also known as Enigma, CLIM1, CLP36, or Elfin, is a 329 amino acid cytoplasmic protein that associates with actin stress fibers at the cytoskeleton. This gene encodes a member of the enigma protein family. The protein contains two protein interacting domains, a PDZ domain at the amino terminal end and one to three LIM domains at the carboxyl terminal. It is a cytoplasmic protein associated with the cytoskeleton. Expressed at high levels in skeletal muscle and heart and at lower levels in colon, small intestine, spleen, lung, placenta, kidney, liver, thymus and pancreas, especially PDLIM1 functions as a cytoskeletal protein that is thought to act as an adaptor, bringing target proteins to the cytoskeleton. Enigma/PDLIM7/LMP1 [D10]
Enigma, or PDLIM7 (PDZ and LIM domain 7) or LMP1 (LIM mineralization protein), is a 457 amino acid protein that localizes to both the cytoplasm and the cytoskeleton. Expressed ubiquitously with highest expression in skeletal muscle, spleen, lung and fetal liver, Enigma is thought to function as a scaffold on which protein assembly can occur. Enigma contains three LIM zinc-binding domains and one PDZ domain through which it may also act as an adaptor, linking various proteins to actin filaments found in skeletal muscle and non-muscle tissues. Additionally, Enigma is directly involved in the two mechanisms of bone formation, namely direct bone formation (embryonic flat bones mandible and cranium) and endochondral bone formation (embryonic long bone development), and may play a role in bone fracture repair.
